Detection principle:
Phosphatidyl serine (PS) is normally located inside the cell membrane, but in the early stage of apoptosis, PS can be flipped from the inside of the cell membrane to the surface of the cell membrane and exposed to the extracellular environment. Annexin-V is a Ca2+-dependent phospholipid binding protein that can specifically bind to PS with high affinity. Annexin-V was subjected to EGFP fusion, fluorescein (FITC, PE) or biotin labeling, and labeled Annexin-V was used as a fluorescent probe. Propidine lodide (PI) is a nucleic acid dye that cannot penetrate intact cell membranes, but can enter cells in the middle and late stages of apoptosis and dead cells, causing the cell nucleus to become red. Annexin V/PI double staining method can detect apoptosis, which can distinguish the early apoptosis cells from the late apoptosis and dead cells.
